Cells are dependent on their niche

The extracellular matrix (ECM) is a complex network of proteins and polysaccharide chains that is secreted by cells. Here you see a simplified view of the in vivo ECM that is a key contributor to the cellular microenvironment, also called the cell niche. The ECM is not just an inert structure. It is a dynamic network in which its components are in direct exchange with resident cells. Thus, the ECM serves specific roles, which contribute to cellular function as well as mechanical properties of tissues.

In vitro, culture conditions are usually extremely simplified with respect to representing the natural function of the ECM. Generally, in vitro cell culture serves a minimalist approach that utilizes single ECM proteins or RGD-containing peptides to stimulate integrin binding and therefore cell adhesion.

denovoMATRIX biomaterials combine specific peptides and polysaccharide chains of the glycosaminoglycan type to recapitulate the key functions of the cell niche in vitro. While the peptides facilitate integrin binding and signaling, the glycosaminoglycans act as growth factor reservoirs and thus support cell adhesion and signaling.

Each cell, whether it is a stem cell or a somatic cell, has different requirements and therefore their own specific niche.

denovoMATRIX technology triples lifetime of FGF2 growth factor

Growth factors are part of the cellular microenvironment and are stabilized by the cell surrounding extracellular matrix (ECM) components such as glycosaminoglycans (GAGs).

As denovoMATRIX technology incorporates GAGs, we can provide benefits for stabilizing growth factors in cell culture media by our functional coatings. We could show an extended half-life of basic fibroblast growth factor (FGF2) in comparison to other cell culture coatings. This stabilization supported the maintenance of stemness and promoted the survival of stem cells, especially when applying restricted feeding regimes.
